ABSTRACT

It makes review on the application of CD-DST on clinical treatment and sci-research of the carcinoma of large intestine from its technological principle and characteristics.It views that as an advanced test technology of medical allergy in vitro, there’s successful report in clinical treatment and concerned scientific study of tumor, however, it’s mainly used in lung cancer and gynecological tumor, the application in the carcinoma of large intestine waits for promotion.

ABSTRACT

AIM: To investigate the effects of recombinant human interleukin-13 (rhIL-13) on the expression of proto-oncogene c-mpl in Dami cells, a human megakaryobiastic leukemia cell line. METHODS: The expression of c-mpl mRNA in Dami cells was investigated with RT-PCR. The expression of membrane-bound protein c-mpl on Dami cells was investigated with ligand combination experiment. RESULTS: In RT-PCR experiment, we found the quantitis of expression of c-mpl mRNA in 25 ?g/L rhIL-13 group increased by 24.8%. In ligand combination experiment, we found quantitis of expression of membrane-bound protein c-mpl in 100 ?g/L rhIL-13 group increased by 28.5% ( P

ABSTRACT

Objective To construct recombinant retroviral vector containing human hepatocellular carcinoma-related gene ANGPTL4 ( angiopoietin-like 4) cDNA and to evaluate antitumor effect of recombinant retroviral vector-mediated human ANGPTL4 gene transfer. Methods ANGPTL4 cDNA was cloned in vitro from human liver cell lines HL-7702 and subcloned into plasmid vector pMSCV and sequenced. High-tiler recombinant retrovirus pMSCV-ANGPTLA and blank retrovirus pMSCV packaged under mediation of lipofectamine infected HepG2 cells in vitro, respectively. Flow cytometry and fluorescence microscopy detected expression of GFP (green fluorescence protein) in HepG2 cells. The expression of ANGPTL4 mRNA in HepG2 cells was determined. Results Recombinant retroviral vector pMSCV-ANGPTL4 was constructed successfully. Titer of recombinant retrovirus pMSCV-ANGPTL4 packaged is 1. 4 ? 106 infective viral grains /ml. Titer of blank retrovirus pMSCV packaged was 1. 5 ? 106 infective viral grains /ml. Positive cell rate of HepG2-ANGPTL4 cells group expressing GFP was 68.45% , and average intensity of fluorescence of HepG2-ANGPTL4 cells group was 31.67 -fold as that of HepG2 cells group. Positive cell rate of HepG2-pMSCV cells group expressing GFP was 77.72%, and average intensity of fluorescence of HepG2-pMSCV cells group was 64. 87 -fold as that of HepG2 cells group. The expression of ANGPTL4 mRNA in HepG2-ANGPTL4 cells group was higher than that in HepG2-pMSCV cells group (154%) and HepG2 cells group( 161%). ABSTRACT

AIM: To establish methods of determing ?-eudesmol in atractylis oil from Ermiao Pills(Rhizoma Atractylodis and Cortex Phellodendri Chinensis) by gas chromatography and determing and comparing the chemical compositions between atractylis oil and poultice of Ermiao Pills by decocting separately and together. METHODS: ?-eudesmol content in Ermiao Pills decocting separtely and together was determined and compared by GC using paeonol as the internal standard substance,and contents of berberine,palmatine,jatrorrhizine in Ermiao Pills decocting separately and together were determined and compared by HPLC. RESULTS: The ?-eudesmol content in the volatile oil of decoction together was more than that of the decoction separately.But there was no remarkable difference.Berberine,palmatine,jatrorrhizine contents of decoction separately were more than that of the together. CONCLUSION: The method of determing ?-eudesmol in Ermiao Pills is established.And it has mutual effect in compatibility with Cortex Phellodendri Chinensis and Rhizoma Atractylodis.

ABSTRACT

AIM:To establish the method for determining oridonin and rosemarinic acid in Donglingcao Tablets(Rabdosia rubescens(Hemsl.) Hara). METHODS:Oridonin and rosemarinic acid were determined by HPLC simultaneously with changing ultraviolet-visible wavelength.Chromatographic condition was composed of ODS-C_(18) column,methanol-03.% phosphoric acid (40∶60),UV detection wavelength of oridonin at 238 nm and of rosemarinic acid at 329 nm. RESULTS:The average recoveries were 97.8% and 96.7% and related standard deviations(RSD) were 1.9% and 2.3%. CONCLUSION:This method can be used to determine simultaneousely oridonin and rosemarinic acid in Donglingcao Tablets.
